- W2896435515 abstract "The polymer PLA, poly (lactic acid), has a range of biomedical applications and its main metabolite is lactic acid, an important substrate for energetic requirements of neurons (1, 2). Producing PLA ultrathin fibers by electrospinning consists in an affordable technique that may contribute to the distribution and up take of lactic acid into the brain, improving neurons functionality. PLA was electrospun and the fibers were coated with gelatin to facilitate the implantation. The produced biodevice was implanted under the dorsal skin of the base of the head of C57BL/6 and APP/PS1 (a transgenic mouse model of Alzheimer's disease, AD) mice, and the animals were treated for 4 weeks. After that, they were transcardially perfused. Brains were fixed and removed, and samples were immunostained for DCX (doublecortin, for immature neurons) and amyloid-β (Aβ) plaques. Once implanted, the degradation of PLA fibers released lactic acid. Consequently, in APP/PS1 animals, in the subventricular zone (SVZ), an important neurogenic niche, the biodevice significantly increased (p < 0.01) the percentage of DCX staining, while in the dentate gyrus of hippocampus, the percentage of Aβ plaques was highly reduced (p < 0.001), compared to the control group. We demonstrated that PLA electrospun fibers have advantageous properties for AD, improving neurogenesis and reducing Aβ plaque load in a APP/PS1 animals. We purpose for the first time that PLA ultrathin fibers may be a remarkable and affordable candidate to act itself as an important tool for AD, establishing the relevance of the material as energy substrate provider for the brain.
