- W2896855117 abstract "Alzheimer's disease (AD) has been associated with the accumulation and deposition of amyloid beta (Aβ) peptides in brain, and immunotherapy targeting Aβ provides high potential for AD prevention. We have used a DNA Aβ42 trimer construct for immunization in wild-type mice and the 3xTg-AD mouse model, and we found a substantial amount of anti-Aβ42 IgA antibodies in the immunized mice. This particular antibody response had not been described previously, and might have an impact on the clearing of amyloid from brain in the immunized mice. 3xTg-AD mice and wild-type mice were immunized with DNA Aβ42 trimer. Humoral immune responses in these mice were compared by ELISA analyzes for the anti-Aβ42 IgG and IgA antibody responses. Epitope analyzes for the antibody binding were performed in parallel ELISA tests. Plaque binding was evaluated with plasma from DNA Aβ42 immunized mice by immunohistochemistry in brains from 3xTg-AD mice. DNA Aβ42 immunization led to strong anti-Aβ42 IgG and IgA antibody responses. No significant differences were found in the overall levels of both antibody isotypes in the immunized Balb/c and B6/129F2 wild-type and 3xTg-AD transgenic mice. The epitope analyses for both antibody isotypes showed broad binding of epitopes across the Aβ1-42 peptide sequence. Anti-Aβ42 antibodies of the IgG and IgA isotype from plasma of the immunized mice were found to bind to plaque in AD mouse brains. The IgG and IgA antibody responses might complement each other in the removal of amyloid from brain in the DNA Aβ42 immunized mice. Further studies in the AD mouse model will be used to evaluate this specific finding. Since we had found strong anti-Aβ42 IgA responses previously in DNA Aβ42 trimer immunized Rhesus macaques as well, the humoral IgA antibody response might have implications for the clinical use of this vaccine as immunotherapy in AD prevention." @default.
