- W2896875781 abstract "Alzheimer's disease (AD) is a global epidemic among the aging population. There are 47 million people worldwide living with AD. Although research efforts have focused on clinical diagnosis, treatment and management, and neuroimaging of patients with AD, much remains unknown. It is critical to understand the clinical progression of AD so that we can discover therapeutic targets and develop solutions to manage AD. However, without the innovation and development of high-resolution, multi-dimensional imaging technologies, understanding the intricate cellular pathology underlying AD will not be possible. Canonically, neuropathologic assessment is based on images captured using pigment precipitation methods such as silver staining or immune-peroxidase staining. Efforts to multiplex these approaches to appreciate sub-cellular features using fluorescent reporters are confounded by highly variable, autofluorescence of the human brain; and increasing the level of multiplexing causes fluorescent spectral overlap, thus convoluting analyses and compromising the rigor of quantitation. Recently, use of elemental reporters in biological assays with mass spectrometry readouts provided an innovative avenue to greatly increase targeted multiplexed analysis at the single cell level. We developed and applied an innovative molecular imaging platform called multiplexed ion beam imaging (MIBI) to quantify subcellular interactions for AD-relevant molecules in archival human tissues specimens in regions AD risk regions (i.e. hippocampus). MIBI is robust in the face of the challenging archival human brain imaging. Using this technology, we have optimized and validated with standard IHC >100 metal-tagged antibodies for simultaneous (40-plex) use with MIBI on human hippocampus, across a range of cognitive status. Specifically, we can now simultaneously determine the structural, immunological and sub-cellular landscape of the human brain, which include: AD-related gene products, cellular anatomy, cellular regulation, homeostasis and injury response, resident immune cells and inflammatory/infiltrating immune responses. These unprecedented multiplexed images quantitatively recapitulate known hallmarks of brain aging (i.e. tauopathy) and provide an opportunity to understand the phenotypic landscape of cognitive decline and resilience. Given this, we believe MIBI will illuminate critical gaps in our knowledge, providing better understanding of disease mechanisms in AD, while highlighting future targets or cellular pathologies for therapeutic development.
