- W2897052659 abstract "Although blood pressure (BP) increase may be accompanied by cognitive decline, it remains unclear what cognitive impairments emerge due to the development of age-associated hypertension. Hippocampal memory alterations have been described in Dahl Salt-Sensitive rats (DSS). DSS develop age-associated hypertension and cardiovascular changes even in the absence of high salt intake. In this study we address the cognitive functions that decline with age-dependent hypertension in DSS vs. normotensive Sprague-Dawley rats (SD). A cross-sectional study using 53 male DSS and SD (3 and 12-month old, n=8-14/group) included BP (plethysmography tail cuff method) and a battery of behavioral tests to assess several components of cognition, i.e., memory, exploratory behavior, and anxiety, and included: vertical activity and distance traveled in an open field test (OFT); balance and motor coordination learning in a rotarod test; memory and object exploration in a novel object recognition test; hippocampal-dependent memory in a version of the Morris Water Maze (MWM) test. 3-mo SD and DSS did not differ in BP, cognitive function, or motor learning tasks. 12-mo DSS, unlike SD, showed a BP increase by 25 mmHg vs. 3-mo DSS (p<0.01) (Table, Fig. 1). 12-mo SD and DSS differed from 3-mo counterparts on OFT in total distance traveled (p<0.0001) and vertical activity (p<0.0001), on the latency during rotarod test with a main effect of strain (p<0.0001), on time exploring objects in the test phase of the novel object recognition task with a main effect of age (p<0.0001) and an interaction of age and strain (p=0.0088) (Fig. 2,3). BP negatively correlated with distance traveled (OFT) in DSS (R=-0.54, p=0.01). 12-mo DSS demonstrated impairment in identifying the location of the invisible platform in a MWM (Table). Observable behavior in aged hypertensive DSS may reflect differences in sympathetic activation, anxiety, hyperactivity, or acceleration of memory impairment; the change in motor function, however, is delayed in DSS vs. SD. Age-associated development of the impairments in several memory components along with the development of hypertension indicates that DSS is useful as a novel model of vascular dementia.
