Inflammation-induced impairment of this process by oxidation of Met476 has been shown to produce dense clots that are resistant to fibrinolysis and pose the threat of thrombosis. To understand the structural basis for the mechanism, we performed enhanced sampling molecular dynamics simulations on the human αC domain dimer in implicit solvent. The only experimental structure for the αC domain is based on the bovine structure so the dimer was modeled using a human homology model. We selected the seven most sampled structures from the trajectories to understand how oxidation affects the stability of the dimer. Methionine oxidation increases flexibility of the αC domain, reduces the size of the hydrophobic core and inhibits dimerization. After evaluating the stability of the minima in explicit solvent, we calculated the interface occupancy of all residues and predicted two dimeric and two monomeric mutants. Met476Gln and Leu478Ala are expected to form monomers by disrupting their wild type contacts. In contrast, Thr486Ala and Met476Val are not expected to significantly change dimerization properties because Thr486Ala displays no occupancy at the interface andMet476Val is a functionally synonymous mutant." @default.
