- W2897375890 abstract "Soft agar assay is an established in-vitro method for assessment of cellular tumourigenicity. Traditionally it involves the microscopic observation of anchorage-independent growth leading to colonies formation after cells are seeded into agar, as evidence of cellular transformation. To compare the assessment of HeLa cell tumourigenicity with single planes versus 3-dimensional z-stack volumes on soft agar assay. A defined number of HeLa cells in culture medium were mixed with agar and layered onto 3 agar plates. The plates were incubated at 37°C and in 5% CO2 in humidified incubator for 3 weeks. The cells were then stained in the plates. Colonies were imaged on single planes and in z-stack volumes of the same area microscopically, and analysed with computer software for the count of colonies, total area of colonies, average size of colonies and percentage of area occupied by colonies. The mean number of colonies and average size of colonies picked up by single planes and z-stack volumes were 16.0 ± 6.1 versus 779.3 ± 444.1 (p=0.04) and 278.9 ± 127.7 versus 19.4 ± 10.7 units (p = 0.03) respectively. The total area of colonies detected were 4957.3 ± 4046.5 versus 12113.0 ± 3052.6 units respectively (p = 0.07). The use of z-stack volumes allows statistically significantly more colonies and those of smaller size to be picked up compared with single planes in soft agar assay. This may increase the accuracy in quantitative in-vitro assessment of tumourigenicity on soft agar assay." @default.
