- W2897400549 abstract "Processing of spatial and object information relies on posterior-medial and antero-lateral temporal systems, respectively (Ritchey, Libby and Ranganath, 2015; see Fig.1). This suggests that tau pathology, which early and commonly accumulates in the temporal lobe in old age, might affect domain-specific memory function differentially. We tested how in vivo measures of tau and Aβ relate to object vs. scene memory function in cognitively normal older adults (OA). Forty-seven OA (78±6yrs) and 21 young adults (YA; 26±4yrs) performed a mnemonic discrimination task developed by Berron et al. (2018) that poses high demands on memory precision and dissociates object vs. scene memory (Fig. 2). High-resolution fMRI data were acquired at 3T while subjects performed the task (1.5mm3 isotropic resolution, whole brain; Fig. 3). In OA, Aβ burden was measured with [11C]PiB PET (23 PIB-, 24 PIB+) and tau accumulation with [18F]AV-1451 PET. Effects of age, global Aβ and regional tau measures in Braak composite regions on domain-specific memory performance and function were tested. OA performed worse than YA across both domains (n.s. group x domain interaction; p>.80) as reported previously. There was no relationship between Aβ (Fig.4, left)and domain-specific memory performance. However, higher AV-1451 SUVRs in Braak I/II (r=0.33, p=.03; Fig.4, right) and III/IV (r=0.27, p=.06) regions were related to relatively worse object than scene memory. Initial analyses using FreeSurfer derived regions suggested higher task activation and less domain-specificity in several regions for OA with higher tau PET measures." @default.
