- W2897533474 abstract "It is becoming more important to prevent the onset of AD through improving lifestyle or diet condition. Probiotics are living microorganisms that are known to confer health benefits onto the host. Surprisingly, some probiotics have been supposed to influence the central nervous system and behavior via the microbiota–gut–axis, raising the possibility that treatment with probiotics could be an effective therapeutic strategy for managing neurodegenerative disorders such as AD. Here, we investigated the effects of oral administration of a probiotic Bifidobacterium strain, B. breve A1, on behavior and physiological processes in AD model mice. The animal model of AD was induced by intracerebroventricular (ICV) injection of Ab. B. breve A1 was orally administrated daily at a concentration of 1×109 cfu starting 2 days before ICV injection. The cognitive function was assessed by Y maze and passive avoidance test at 7 days after ICV injection. Brain samples were obtained after behavioral test, and gene expression in hippocampus was analyzed by RNA-sequencing. Mice with ICV injection of Aβ showed a significant reduction of alternation behavior in the Y maze test and latency time in the passive avoidance test compared with sham-operated mice, which indicated that Aβ injection induced cognitive dysfunction. We found that administration of B. breve A1 to AD mice reversed the impairment of cognitive function in both working memory and long-term memory. Gene profiling analysis revealed that consumption of B. breve A1 suppressed the hippocampal over-expressions of inflammation and immune-reactive genes induced by Aβ. We also demonstrated that non-viable component of the probiotics or acetate, which is a metabolite of Bifidobacterium, partially ameliorated the cognitive decline. The present study demonstrated that the effect of B. breve A1 in the improvement of cognitive function in this AD model mouse and the possible mechanism via modulating the immune response and neuronal inflammation. These results suggest the potential of B. breve A1 for preventing cognitive impairment in Alzheimer's disease. Clinical data from pilot study on subjects with mild cognitive impairment aiming to explore the effect of B. breve A1 supplementation on dementia prevention will also be discussed." @default.
