- W2897540529 abstract "Without intervention Alzheimer's disease will devastate public health resources and overwhelm socioeconomic frameworks on a global scale. It is imperative to mitigate this disease with a highly impactful treatment, or a delay in onset. To achieve such goals, we must pool global resources and expertise to generate data that can improve disease understanding and inform design of therapeutic trials at all stages of the disease. It is within this context that CONCORDE-AD (Connecting Cohorts to Diminish Alzheimer's Disease) was created: to foster scientific exchange, generate insights and contribute to the evolving clinical science in AD. The current network includes longitudinal cohorts that have been extensively characterized both clinically and biologically. The collaboration model involves sharing of independently conducted analyses addressing key questions about disease progression, biomarker-clinical correlations and patient-relevant outcomes. This model facilitates evaluations into the impact of disease heterogeneity (e.g. geographic and cultural variations, differences in healthcare systems, study inclusion/exclusion criteria) on critical scientific questions. CONCORDE-AD comprises seven cohorts across Australia, Europe and North America representing data from >20,000 individuals across the disease spectrum - from cognitively unimpaired to AD dementia as well as other dementias - with follow-up of up to 22 years. The network includes the Australian Biomarker and Lifestyle Study of Aging (AIBL), Swedish BioFINDER Study, Personnes Agées QUID (PAQUID), 3-City Study Bordeaux, AMI cohort of elderly farmers, Baylor Alzheimer's Disease and Memory Disorders Center, and the Mayo Clinic Study of Aging. Evaluation of all available data from each cohort allowed study comparisons and recommendations for the ‘core dataset’ essential to facilitate combined analyses. Although each cohort was established with different objectives, the opportunity to combine data from seven cohorts provides a unique resource to replicate findings, explore causes of heterogeneity, increase sample size/power, and leverage disease area expertise across different settings. Expansion of the network to include other well-characterized cohorts and eminent investigators would further strengthen the potential to better understand, and eventually conquer, Alzheimer's disease." @default.
