- W2897612576 abstract "Brain volume loss is a marker of dementia-associated neurodegeneration. We investigated the relative contributions of aging and seven modifiable dementia risk factors (MDRF, hypertension, diabetes, obesity, frequent alcohol consumption, smoking, inadequate sleep, and lack of college degree education) for explaining differences in brain volume. Data from 8312 UK Biobank participants (62.4±7.4yrs, 47.1% men) without brain neurological or psychiatric diseases who underwent 3T structural brain MRI (Access Agreement 18545) were used to examine effect sizes and anatomical distributions of brain grey and white matter differences associated with MDRF relative to those found with aging. A multiple regression model estimated independent contributions of age and MDRF (corrected p<0.05). Brain volumes were 0.41% lower mean volume/year of age, with greater age-related differences in grey matter (0.53%) than in white matter (0.24%). MDRF accounted individually for variable additional, independent differences in brain volume (all values expressed as % lower total brain volume (TBV)): diabetes (2.1%) > frequent alcohol consumption (0.68%) ∼ obesity (0.51%) > hypertension (0.31%). An association between smoking and total grey matter volume was found, but subjects’ reports of inadequate sleep or lack of college degree education were not significantly associated with brain volume measures. Voxel-wise parametric variations tested using Statistical Parametric Mapping (SPM 12) showed 74% of TBV varied negatively with age. This included 84% of the total grey and 59% of the total white matter volume (p<0.025, family-wise error [FEW] corrected). Focal brain volumes negatively associated with the seven MDRF (all values expressed as % TBV) were within the brain volume influenced by age: diabetes (23.1%) > obesity (6.2%) ∼ hypertension (5.6%) > frequent alcohol consumption (2.3%) > lack of college degree education (0.8%) > smoking and inadequate sleep (<0.01%) (p<0.025, FWE corrected). Our results highlight highly significant associations between lower brain volumes and MDRF. The greatest effect size was found with diabetes, followed by hypertension, obesity, and frequent alcohol consumption. This suggests the potential relative impacts on late life cognitive impairment from public health measures targeting these risks. The anatomically overlapping independent aging and MDRF effects suggest that they influence common brain regions additively." @default.
