- W2897618881 abstract "Neuropathological hallmarks of Alzheimer's disease (AD) include amyloid-β (Aβ) plaques and neurofibrillary tau tangles in brain. Diacylglycerol (DAG), which is a docosahexaenoic acid (DHA) precursor, has been shown to interact with Aβ and influence tau phosphorylation. Moreover, DAG was found in one study to show an increase in a specific DAG species in AD and amnestic mild cognitive impairment (MCI) patients, when compared to controls. Genetic mechanisms underlying potential DAG contributions to AD are unknown. To better understand how Aβ and tau are associated with DAG pathway-related genes, we performed a gene-based association analysis using three CSF biomarkers (Aβ1-41, total-tau, p-tau) for AD, which reflect AD pathology. DAG pathway-related candidate genes (n=172) were manually curated from existing databases (Human Metabolome Database (HMBD), LPID Metabolites and Pathways Strategy (LIPID MAPS), and STRING). Participants from the Alzheimer's Disease Neuroimaging Initiative (ADNI) with CSF measures and genotype data were included. The genotype data were imputed using the HRC r1.1 reference panel after standard sample and SNP quality control procedures. Next, CSF amyloid beta (AB), phosphorylated tau (p-TAU), and TAU data was obtained from the ADNI-1 database (http://adni.loni.usc.edu). GWAS was performed using linear regression under an additive model in Plink http://pngu.mgh.harvard.edu/wpurcell/plink/. Age, sex and APOE were used as covariates. A gene-based association analysis was performed using an extended Simes (GATES) procedure implemented through Knowledge-Based Mining System for Genome-wide Genetic Studies (KGG; http://grass.cgs.hku.hk/limx/kgg/) on each SNP-based association result. Targeted gene based association analysis for DAG-pathway related candidate genes identified PNPLA2 (p-value=6.55E-05) as significantly associated with CSF t-tau and p-tau levels after adjusting for multiple testing using the Bonferroni correction. In addition, AKAP13 (p-value=6.40E-04) and LIPF(p-value =5.50E-04) were marginally associated with CSF Aβ and p-tau levels, respectively. A DAG pathway-related gene PNPLA2 was associated with CSF t-tau and p-tau levels. PNPLA2 (patatin-like phospholipase domain containing 2) functions in lipid metabolism and signaling with genetic roles in autism and physiological contributions to aging. AKAP13, which was marginally associated with Aβ levels, is known to phosphorylate tau. Studies of DAG metabolite species are planned and replication of the genetic associations will be important." @default.
