- W2897645312 abstract "To perform precise endocrinian functional assays, we have tried to set up an analytical method of 17-oxosteroids (17-OS) and 20-hydroxy-21-desoxysteroids from urine by gas-liquid chromatography (GLC). These steroids have been chosen for their interest in clinical chemistry with the aim to do an assay fulfilling the best qualitative and quantitative performance while remaining relevant to the daily requirement of a clinical laboratory. This study has been made possible because we have the use of a mass spectrometer coupled to GLC (GLC-MS) which allowed us to identify each Chromatographic peak, especially androsterone, 11β-hydroxy-androsterone, 11βhydroxy-etiocholanolone, pregnandiol-20β, pregnandiol-20α, pregnanetriol-20β and pregnanetriol-20α. We have particularly studied a fast procedure leading to the complete formation of stable trimethylsilyl derivatives. We have also looked for the best conditions for separation and identification of these 12 catabolites through the variation of the analytical parameters (isotherm, temperature programmation, carrier gas flow, etc.) and by using different liquid phases (OV-1, OV-17, SE-60, Dexil-300). Finally, we have kept the two liquid phases OV-1 and OV-17, which are compatible with the working temperatures and complementary by their separating pattern.
