- W2897771808 abstract "During aging and under the influence of stress amount of injured pyramidal neurons in hippocampus is increased. It's believed, decreased functional activity of serine racemase in neurons of the brain promotes reduction of learning ability. 12- and 24-month-old rats were subjected to seven days combined stress in a special chamber (6 isolated compartments of equal volume) with removable multi-modal stressors. We evaluated the neuropsychiatric state of animals and determined the degree of neuronal injury and expression of serine racemase (SR) in the hippocampus. In 24-month-old rats motor and orienting-exploratory activity decreased in comparison with the 12-month-old rats. Combined stress impact equally reduced motor and orienting-exploratory activity of rats in both groups. In 24-month-old rats after stress exposure cognitive deficits was observed, it manifested in the deterioration of the formation and preservation of the memory trace. In groups of 12- and 24-month-old rats after combined stress pericellular edema, vacuolization of cytoplasm in perikarya of neurons were found, the share of pyknotic damaged hyperchromatic pyramidal neurons increased in CA3 (p<0.05), the relative density of pyramidal neurons was decreased in CA3 (p<0.05), expression of SR in dendrites in radial layer of CA1 and CA3 was decreased. Cognitive impairments more prominent in aged rats (24-month-old) exposed to combined stress. Development of amnesia, significant increase of dementia manifestations after combined stress are associated with increased number of injured pyramidal neurons, decreased relative density of neurons in the pyramidal layer of hippocampus, because the hippocampus is involved in spatial orientation, emotional reactions including to stressful stimuli. Age-related decreased expression of SR in dendrites of radial layer of CA1 and CA3 reduces learning ability and can be regarded as a sign of decreased NMDA-R dependent synaptic plasticity. The observed morphological and functional changes indicate more prominent emergence of neurodegenerative processes after combined stress in aged rats." @default.
