- W2897869940 abstract "Olfactory deficits are prevalent in early AD. However, it is unclear how this sensory deficit relates to AD-related cognitive decline and dementia. We have shown that the default mode network (DMN) was strongly deactivated during odor processing in normal human subjects, suggesting that the formation of an odor percept may be drawing cognitive, attentional, and memory resources [1]. Many other studies have already shown profound DMN involvement in AD. To establish the basis for using olfactory deficits as a diagnosis marker for AD, this study investigated the relationship between the DMN and olfactory network (ON) in AD. 31 healthy control (HC, mean age= 69.5 years, 15 F), 19 mild cognitive impairment (MCI, mean age= 72.8 years, 10 F) and 12 AD (mean age= 73.7 years, 8 F) subjects completed an odor-visual association fMRI paradigm at 3T in Figure 1. The participants’ smell function were evaluated using the University of Pennsylvania Smell Identification Test (UPSIT). Independent Component Analysis (ICA) was used to interrogate the interplay between the DMN and ON in respective cohorts [1]. The UPSIT and DRS-memory scores in AD, MCI and HC groups were significantly different among groups at a p value of <0.01. ICA maps in Figure 2 showed markedly less fMRI activation in MCI and AD groups following a descending trend of the UPSIT scores. A striking deterioration in the DMN suppression was demonstrated in MCI and AD (Figure 3). In MCI, the effective connectivity (EC) between the DMN and ON was disrupted." @default.
