- W2897880463 abstract "The epsilon4 allele of Apolipoprotein E (APOE) has been showed to be associated with dementia in particular Alzheimer's disease (AD). Many studies have suggested a low frequency of epsilon4 allele in sub-groups of Indian population. In this study we determine the frequency of epsilon4 and its association with AD in study population from South India. 133 subjects (healthy controls, subjects with mild cognitive impairment-MCI and AD) residing in the two South Indian cities of Bengaluru (N=98) and Hyderabad (N=35) who are part of a follow-up study were included for this work. The two APOE single nucleotide polymorphisms rs429358 and rs7412 which determine the APOE epsilon allele status were genotyped using Sanger sequencing. For association analysis only unrelated individuals were retained, first and second degree relatives were excluded. For association with MCI/AD status, subjects who carried at least one copy of the epsilon4 allele were compared with non-carriers of the allele. The allele frequency of epsilon4 was 0.15, epsilon3 was 0.80 and epsilon2 was 0.05 in this South Indian study population. After excluding subject with MCI and AD the frequency of epsilon4 was 0.14, epsilon3 was 0.80 and epsilon2 was 0.06. This allele frequency of epsilon4 in the Indian population is much higher than reported by previous studies on dementia. The epsilon4 allele was associated with MCI/AD status in subjects from Hyderabad (OR= 11.50, 95%CI=2.06-64.34, P=0.004) and showed a non-significant trend for association with MCI/AD in the subjects from Bangalore. The epsilon4 allele of APOE showed a high allele frequency in this study compared to previous reports on dementia studies from India and was associated with MCI/AD status." @default.
