- W2897909969 abstract "Background Human periodontal ligament stem cells (hPDLSCs) are similar to bone marrow mesenchymal stem cells and differentiate in vitro into osteoblasts, chondrocytes and adipocytes. 1,25(OH)2D3 are known to promote osteogenic differentiation of hPDLSCs under physiological conditions. Aim/Hypothesis The aim of this study was to investigate these effects under inflammatory conditions. Material and Methods HPDLSCs of six donors were treated with different concentrations of 1,25(OH)2D3 (0.1–10 nM) in the presence and absence of Porphyromonas gingivalis (P.g.) lipopolysaccharide (LPS) (1 µg/mL) or TLR2 agonist Pam3CSK4 (1 µg/mL) for 48 hours. Treatment of hPDLSCs with P.g. LPS or Pam3CSK4 alone was performed as control. Gene expression of osteocalcin (OC), osteopontin (OPN) and Runt-related transcription factor 2 (RUNX2) was measured by qPCR. Results 1,25(OH)2D3 induced significant expression of OC and OPN in hPDLSCs in a concentration dependent manner. Highest tested concentrations of 1,25(OH)2D3 induced increase of OC and OPN expression by about 14 and 3 times, respectively. In the presence of Pam3CSK4, 1,25(OH)2D3 at similar concentration induced only about 7-fold increased expression of OC, which was significantly lower (P < 0.05) compared to normal conditions. Similarly, in the presence of Pam3CSK4, the enhancement of OPN gene expression by highest tested 1,25(OH)2D3 concentration was only about 1.5 times but no significant difference compared to control conditions was found (P = 0.17). The effect of 1,25(OH)2D3 on expression of OC and OPN was not altered in the presence of P.g. LPS. No significant effect of vitamin D on RUNX2 expression was observed. Conclusions and Clinical Implications Our data showed that the effect of vitamin D on the expression of osteogenesis-related factors in hPDLSCs is altered under inflammatory conditions. The effectiveness of vitamin D on local bone metabolism might be diminished in individuals with inflammatory disease." @default.
