- W2897922721 abstract "We link the following knowledge for monocytes, amyloid beta (Aβ) and docosahexaenoic acid (22:6n-3; DHA) in our current initial study: (1) Microglia are myelogenous brain resident innate immune cells, and are main and early responders in the central nervous system (CNS) immune defense. They also play a role in CNS Aβ clearance, via phagocytosis and degradation, and in upholding of synaptic homeostasis. (2) Peripheral myeloid cells, such as monocytes and macrophages, are regulated in parallel to the microglia histiocytes in many respects and share phagocytic properties. In addition, these cells may circulate to and infiltrate the CNS and they have been suggested to play a role in AD pathogenesis such as cerebral amyloidosis. (Wisniewski et al. 1989, Wisniewski et al. 1991) (3) Long chain polyunsaturated fatty acids (LCPUFAs) are found in significant amounts in the brain and their incorporation in cell membranes makes them critical for a range of functions. More specifically, DHA is a promising LCPUFA-nutraceutical proposed to inhibit AD development. Mechanisms for this putative inhibition are so far undetermined. We used monocytic THP-1 cells as model system and IP nLCMS as analytical method to investigate DHA's effects on monocytic Aβ40 processing. In the cell lysate intact Aβ40 was sparsely detected, whilst Aβ40 degradation products were widely detected proving both monocytic engulfed and degraded Aβ40. We also semi- quantitatively compared the Aβ40 peptides for the conditions and these comparisons imply that DHA functions as a catalyst for both monocytic phagocytosis and catabolism of Aβ40. These Aβ cell culture degradation patterns both harmonize with in vitro experiments for lysosomal degradation (Rogeberg et al. 2014) and that obtained from patient harvested monocytes (AAIC poster presentation, Torsetnes et al. 2018).
