- W2897962380 abstract "The adipocyte-derived hormone leptin has been discussed to exert beneficial effects on brain-structure and cognition. Leptin potentially promotes neuronal survival, especially in regions with high leptin-receptor density, such as the hippocampus(Hawrylycz2012;Holden2009;McGregor2017). However, higher leptin values are not per se protective. In obesity and inflammatory states chronically elevated leptin levels are a marker of leptin-resistance, which in turn poses a health risk(Iikuni2008;Martin2008). Only few studies account for possible effects of obesity-associated leptin-resistance and investigate effects of leptin on the hippocampus and assumed implications for cognition(Witte2016). Therefore, we combined anthropometric measures, high-resolution MRI with cognitive measures and investigate the effects of leptin on hippocampus volume and cognitive performance in a large cohort of healthy humans. Subjects: 1644 healthy individuals from the Leipzig LIFE-Adult-Study (mean-age=57.99±15.16years (range=19-82); mean-BMI=27.11±4.5kg/m2(range=17-55). Exclusion: stroke, major brain-pathologies, central-nervous medication Neuroimaging of 3Tesla high-resolution T1-weighted images: Hippocampus volumes were estimated using automatic segmentation analysis implemented in FreeSurfer5.3.0. Statistical analysis: Linear regressions(enter model): with independent variables (age, sex, leptin(sex-standardized), waist-hip-ratio(sex-standardized), dummy-overweight, dummy-obese, leptin*dummy-overweight, leptin*dummy-obese) and dependent variables(left/right hippocampus volume, verbal learning, semantic and verbal fluency, trailmaking-task. Linear regressions showed significant positive relationships between leptin and left(p=0.006;standardized-B=0.14) and right(p=0.006;standardized-B=0.14) hippocampus volume for the whole sample. Furthermore, lower age(p<0.001;standardized-B<-0.42), being female (p=0.001;standardized-B=0.08) and lower WHR(p=0.01;standardized-B<-0.07) were predictive of higher left and right hippocampus volume in the same models. The interaction-terms were not significant. Leptin was not predictive for cognitive performance in this sample. In another independent sample(n=53), left and right hippocampus volume derived from ultra-high-field anatomical MRI at 7T also showed significant correlations with leptin in older non-obese individuals after correcting for age, sex and percent body fat(both p<0.02)." @default.
