- W2897967103 abstract "Early and accurate diagnosis of Alzheimer's disease (AD) is limited by costly or invasive methods to assess amyloid accumulation. Event related potentials (ERPs) may be a potential alternative biomarker of brain dysfunction attributable to amyloid accumulation. ERP peak latencies and amplitudes differ between mild AD patients and healthy older adults (Cecchi et al., 2015), and between patients with mild cognitive impairment (MCI) due to AD versus those with MCI due to other etiologies (Green et al., 2015). In this preliminary study, we investigate the utility of ERPs as a potential biomarker of the brain dysfunction due to Alzheimer's disease. Twenty-seven subjects who presented with memory loss underwent standard clinical workup including history and physical, neuroimaging studies, and a neuropsychological battery, leading to a clinical diagnosis. All subjects underwent a florbetapir positron emission tomography (PET) scan to assess amyloid status and consented to an ERP session using a three-tone auditory oddball paradigm. Individual ERP measures between PET+ (n=11) and PET- (n=16) groups were analyzed using receiver operating characteristic (ROC)-curves. ROC analysis was used to determine cutoff values and to assess maximal sensitivity and specificity of ERP measures against the PET status (PET+/-). Of the 36 ERP measures acquired, the P50 peak amplitude for standard tones (P50 std amp: p=.040, sensitivity (se)=.64, specificity (sp)=.69), P50 average amplitude to target tones (P50 tgt avg: p=.041, se=0.73, sp=0.75), P200 peak amplitude (P200 tgt amp: p=.025, se=0.67, sp=0.66), and average amplitudes for target tones (P200 tgt avg: p=.023, se=0.91, sp=0.63) displayed sensitivity and specificity values that hold the promise of clinical utility in the future." @default.
