• W2898035074 abstract "Prenatal screening for aneuploidy via next-generation sequencing of cell-free DNA (cfDNA) in the maternal circulation is the current standard of care for advance maternal age. Circulating cfDNA is derived from both the mother (primarily maternal hematopoietic cells) and the fetal-placental unit (primarily outer placental cells, the synctiotrophoblast). Despite advances in laboratory technologies, a low fetal fraction of cfDNA results in assay failure. Factors known to systematically reduce fetal fraction include early gestational age and maternal obesity. The aim of this research is to explore whether placental volume at the time of cfDNA collection correlates with cfDNA fetal fraction. Retrospective chart review study from 1/2016 to 12/2017. We identified 28 patients with cfDNA assay failure and 281 patients with cfDNA assay results. For each patient, we collected available data for gestational age, Crown–rump length, placental volume and cfDNA fetal fraction. Out of 28 patients with cfDNA failure, 23 had placenta volume, nuchal translucency and Crown–rump length data. Independent sample T-tests were used to compare placenta volume, Crown–rump length, and nuchal translucency between the cfDNA assay failure group and cfDNA assay result group. The mean placental volume for cfDNA assay failure group was 61.18 mm2(range: 27.7 mm2, 90 mm2; SD: 19.03), and the mean placental volume for cfDNA assay result group was 62.65mm2(range: 24.6 to 97.9 mm2; SD: 18.70 mm2) p value of 0.72. The mean nuchal translucency for cfDNA assay failure group was 1.46 (SD: 0.335) and for cfDNA assay result group was 1.58 (SD: 0.439),p value of 0.2. The mean Crown–rump length for the cfDNA assay failure group was 58.22 (SD: 7.82), and for the cfDNA assay result group was 60.99 (SD: 9.19),p value of 0.183. Our data did not show a significant difference in placental volume, nuchal translucency, and Crown–rump length between the cell free DNA result group and cell free DNA assay failure group.
