- W2898883705 abstract "Most mitochondrial proteins are synthesized as preproteins in the cytosol and subsequently imported into the organelle. We are interested in the process of mitochondrial import of Peroxiredoxins (Prxs), which are thiol-dependent peroxidases. We have previously showed that yeast mitochondrial Prx1 contains a bipartite mitochondrial targeting peptide, consisting of a positively charged N-terminal presequence followed by a hydrophobic segment. While the presequence promotes Prx1 sorting into the matrix, the hydrophobic segment induces the lateral release of Prx1 preprotein into the lipid phase of the inner membrane. The hydrophobic segment is then removed by inner membrane peptidase, which releases the protein into the intermembrane space. The lateral release of preproteins containing a bipartite signal is controlled by Mgr2. This protein interacts with the preproteins in transit through the inner-membrane translocase TIM23 and delays their lateral release into the inner membrane. In this study, we performed the submitochondrial localization of Prx1 in WT and ΔMGR2 yeast cells. As expected, the lack of Mgr2 increased the lateral release of Prx1 into the inner membrane as confirmed by a higher level of Prx1 in the intermembrane space in the ΔMGR2 cells compared with WT cells. We intend now to study the effects of Mgr2 overexpression on Prx1 submitochondrial localization and the oxidative stress response of ΔMGR2 cells. In parallel, we are investigating the molecular mechanisms of mammalian mitochondrial Prxs import and submitochondrial localization. Preliminary results indicated that mammalian mitochondrial Prx3 from HEK293 cells also have dual mitochondrial localization like yeast Prx1. These results indicates that the targeting mechanisms of mitochondrial Prxs are evolutionary conserved. The identification of the proteins involved in targeting Prx3 to distinct sub-mitochondrial compartments is underway and it will be important for understanding the role of these peroxidases and its association with mitochondrial dysfunction in diseases." @default.
