- W2898987577 abstract "Horse dent corn (Zea mays var. indentata) hydrolyzed by protease enzyme of Rizophus oligosporus strain C1 has potential use as fortificant of natural folic acid. Purification of target components by means of ultrafiltration (UF) membrane (20000 MWCO) equipped in dead-end stirred filtration cell (DESFC) was conducted on 2 (two) kinds of yellow corn hydrolysate (YCH) and white corn hydrolysate (WCH) at room temperature, stirrer rotation speed (SRS) 200, 300 and 400 rpm, and transmembrane pressure (TMP) 20 and 40 psia for 30 minutes. The experiment result showed that interactions amongst types of treatments affects on both composition and folic acid dominationof folic acid monomer. Based on optimization of folic acid, the best treatment on YCH and WCH were achieved at operation condition with SRS 200 rpm and TMP 40 psia. In this optimum condition, UF membrane was able to separate target components in both retentates of YCH and WCH, such as folic acid 2.64 % and 17.08 %, total solids 64.13 % and 54.26%, reducing sugars 81.21 % and 32.10 %, total sugars 25.64 % and 8.87 %, dissolved protein 150 % (1.5 times) and 13.79 %, and total acids 17.5 % and 97.64 %, respectively compared with each component before process (feed). In this optimum condition, it is generated both concentrates of YCH and WCH with compositions of folic acid 89.79 µg/mL and 114.96 µg/mL, total solids 24.58 % and 28.37 %, reducing sugars 0.37 mg/mL and 0.55 mg/mL, total sugars 273.14 mg/mL and 358.08 mg/mL, dissolved protein 1.00 mg/mL and 0.66 mg/mL, and total acids 0.235 % and 1.37 %. Identification on folic acid monomer in both concentrates of YCH and WCH were dominated by monomer having molecular weight (MW) 442.27 Dalton (Da.) and 442.58 Da. with relative intensities 60 % and 100 % at mass spectra m/z 441 – 444." @default.
