- W2899100199 abstract "Objectives: Telomeres are repeated sequences at the end of chromosomes responsible for preventing DNA degradation and unnecessary recombination. Shorter telomeres are associated with increased risk of hypertension in humans. Angiotensin converting enzyme inhibitors (ACEi) may promote telomerase activity, responsible for maintaining telomere length. We aim to investigate the effects of ACEi treatment on telomere length and associated genes in renal cortex and heart ventricle of spontaneously hypertensive rats (SHR). Methods: Male SHRs were treated daily with ACEi perindopril (0.1 mg/kg/d) during 10 to 14 weeks of age. Tail cuff systolic blood pressure (SBP) was measured at 14-and 20-weeks old. Tissues were collected at 10-, 14- and 20-weeks (n = 5 per group). Telomere length was reported as a T/S ratio and measured along with mRNA expression using real-time quantitative PCR. Results: After perindopril treatment telomeres were longer at 14- (FC = 1.13, P < 0.05) and 20-weeks old (FC = 1.1, P < 0.05) in the cortex. In the ventricle, telomere length was reduced at 20-weeks old (FC = 0.95, P < 0.05). SBP was lower in the SHR at 14-weeks old (162v206mmHg, P < 0.001) and remained reduced at 20-weeks old (177v220mmHg, P < 0.05). Telomere associated genes were differentially expressed at 14- (Tert FC = 1.7, Pnuts FC = 1.5, all P < 0.05) and 20- (Terc FC = 1.8, and Pnuts FC = 1.9, all P < 0.05). Conclusion: Our results suggest that early ACEi treatment influences SBP and telomere length in the SHR. These findings indicate that epigenetic regulation of telomere length by ACEi is associated with long-term protection from hypertension." @default.
