- W2899200883 abstract "Objectives: Chronic intermittent hypoxia (CIH), a feature of obstructive sleep apnea, enhances carotid body (CB) discharges, leading to sympathetic overflow and hypertension. The CB ablation regularized arterial blood pressure (BP) during CIH, showing that the enhanced CB chemosensory drive to the brainstem cardiovascular control centers is required for the hypertension. CIH produced systemic and local CB oxidative stress, which contribute to enhance the chemosensory discharge. Thus, it is plausible that brainstem oxidative stress may contribute to the hypertension. To assess the contribution of brainstem oxidative stress to the hypertension, we compared the effects of intracerebroventricular (ivc) Tempol infusion with the effects of CB ablation in CIH-hypertense rats. Methods: Sprague-Dawley rats (200 g) were exposed to CIH (5% FiO2, 12 times/h, 8 h/day) for 28 days and BP was measured by telemetry in conscious animals. At 14 days, under isoflurane anesthesia, rats were implanted with mini-osmotic pumps to infuse continuous Tempol ivc (60 μg/ kg day), and rats were keep on CIH for another 14 days. Results: CIH increased mean BP from 93 ± 0.9 mmHg to 103.1 ± 0.6 mm Hg in 3–4 days (n = 8 rats). Infusion of Tempol icv slightly reduced the elevated BP to a mean BP of 100.3 ± ,0.8 mmHg (P > 0.05) in 10–14 days. On the contrary, the ablation of the CBs promptly totally restored the normal BP in one day, effect that is maintained despite of the presence of CIH. Conclusion: Our results indicate that CB plays a crucial role in the hypertension induced by CIH since CB ablation completely restored the elevated BP, while the administration of Tempol icv did not restore the baseline BP.
