- W2904598706 abstract "Abstract Negative-strand RNA (NS RNA) viruses, which may have segmented or nonsegmented genomes, are to be blamed for plentiful of grave viral diseases such as measles, rabies, influenza, Crimean-Congo hemorrhagic fever, Ebola, and Lassa fevers in both humans and animals. Their multipotent enzymes called RNA-dependent RNA polymerases (RdRps) transcribe and replicate the viral genome in the host. Thus, viral RdRps are an attractive antiviral drug target as they fulfill the utmost functions performed in viral replication and transcription. Recent and upcoming disease management approaches involve targeting the viral genes through siRNA (small interfering RNA, also known as short interfering RNA or silencing RNA) technology, novel drug delivery approaches to ameliorate the effectiveness of previously approved FDA drugs, and acquisition of novel approaches to developing host immunity against these viral diseases through largely targeting the viral RdRps. Nonetheless, considering the fact that viruses develop resistance against monotherapy using a single drug, one of the appealing options that come up is chemotherapy with the appropriate combination of drugs affecting varied viral proteins or different factors responsible for viral pathogenicity. In this chapter, we have discussed the structural features of endonuclease domain of N-terminal region of L-proteins along with the comparisons between different members of segmented negative-strand (SNS) and nonsegmented negative-strand (NNS) RNA viruses. This chapter summarizes the insights gained from recent structural and functional studies on NS RNA viruses and its polymerase. Furthermore, antiviral strategies are also discussed which particularly focus to combat viral diseases through targeting viral polymerases which can come up as the potential antiviral drug that can reach clinics." @default.
