- W2912187389 abstract "Objective: To develop an ethical and cost-effective animal model which allows human-equivalent neuroimaging that can be combined with invasive neuroscience techniques for investigation of human diseases. Background: Magnetic resonance imaging brain assessments have become a standard method for studying human brain development, particularly in the crucial periods of development from adolescence to early adulthood. However, the standard animal models for brain development have significant limitations due to smaller brain volumes and structural disproportionality, whereas non-human primates with more similar cortices have financial and ethical constraints which limit utility in basic neuroscience research. Therefore, a porcine model ( Sus scrofa domestica ) that lacks the limitations of previous models was tested for translational neuroimaging studies of adolescent brain development. Design/Methods: Twelve three-month old Sinclair miniature pigs were longitudinally evaluated during adolescent development with MRI scans every 25 ± days using a 3-Tesla scanner. DWI protocols were analyzed with diffusion kurtosis and bi-exponential modeling providing for measurements of fractional anisotropy (FA), kurtosis anisotropy (KA), and permeability-diffusivity index (PDI). Results: Significant longitudinal effects of brain development were observed for whole-brain average FA, KA, and PDI (P Conclusions: These changes are evidence for ongoing myelination and maturation of porcine cerebral white matter with regional differences which mirrored human development, thus validating the porcine model without the limitations of previous models. This porcine model can be used in human scanners and imaging protocols, providing a direct translational impact, and can additionally be complemented with invasive neuroscience techniques.
