- W2912297294 abstract "Ca2+-triggered neurotransmitter release by synaptic exocytosis is a highly regulated process for neuronal communication. This process depends on several proteins including SNAREs, Munc proteins and complexins and is tightly controlled by Ca2+ sensor proteins. Synaptotagmin-1 (Syt1) is a calcium sensor protein that regulates synchronous neurotransmitter release. While the role of Syt1 in living cells has been described in detail, it remains a challenge to dissect the contribution of membrane remodelling by its two cytoplasmic C2 domains (C2AB) to the Ca2+-secretion coupling mechanism. Here, we study membrane remodeling using pairs of optically-trapped beads coated with SNARE-free synthetic membranes. We explore membrane remodeling in the presence of two proteins: Syt-1 and Doc2b, a Ca 2+ sensor protein that triggers spontaneous neurotransmitter release. We find that the soluble C2AB domain of Syt-1 strongly affects the probability and strength of membrane-membrane interactions in a strictly Ca2+- and protein-dependent manner. When comparing symmetrical (both sides) and asymmetrical (one side) presence of protein on the membranes, Syt1 favors an asymmetrical and Doc2b a symmetrical configuration, as inferred from higher tether probabilities and break forces. This difference is reminiscent of the physiological configuration of the proteins. Further, we quantify the probability of hemifusion in the presence of Syt-1 vs Doc2b, and find that Doc2b allows both hemifusion and membrane bridging, while Syt-1 only bridges membranes and does not allow hemifusion. Further, we reveal lipid composition dependence of membrane remodeling by the calcium sensor proteins. To understand how these proteins affect membranes, we probe membrane mechanical properties using a novel AFM-based method and reveal differences in membrane bending moduli following protein binding. Our approach can be readily extended to explore other fusion-related proteins." @default.
