- W2912348240 abstract "Metals are both essential for cardiovascular health and among the oldest known cardiovascular toxicants. Recent findings, such as large prospective epidemiological studies or finding that metal chelation therapy reducing cardiovascular events in cardiovascular disease (CVD) patients, strengthen the link between metal exposures and disease promotion. However, pathogenic mechanisms for metals promoting human CVD remain poorly understood and are often controversial due to difficulty in interpreting and translating results from high dose animal toxicity studies to causal disease etiologies in humans. There is increasing appreciation that lower, chronic environmental exposures, exposures occurring in critical developmental windows, or exposures at occupational and therapeutic levels may have subtler effects in causing or modifying CVD. Recent epidemiological studies and studies in genetic rodent models that focus on environmental exposures indicate that pathogenic effects of metals on cell signaling and epigenetic regulation are not random and cannot be attributed to competition with endogenous ions or excessive oxidant-mediated injury. Resolution of the molecular understanding of pathogenic metal effects remains a challenge complicated by complex differential concentration and temporal exposure relationships, as well as genetic determinants. This chapter presents pathogenic effects of metals in the heart and vasculature with an emphasis on the cellular and molecular actions underlying CVD promotion and clinically significant toxicities." @default.
