- W2912621289 abstract "Field emission scanning electron microscope (FESEM) provides a number of advantages for detailed assessment of cell and tissue architecture. This chapter focuses on some of the procedures for preparing samples for FESEM imaging under non-cryogenic conditions and the kind of information that can be obtained from studies of mammalian cells in culture. With the ability to identify molecular complexes and the application of backscattered electron detection to localise proteins labelled by immunogold techniques, FESEM offers opportunities to make comprehensive analyses of normal cell and tissue architecture and of the causes and consequences of pathological processes. Immunogold labelling applied directly to samples for examination by backscatter electron detection in the SEM allows exploration of the location and distribution of surface antigens on individual cells. Application of immunogold labelling procedures following demembranation allows identification of molecular components of, for example, cytoskeletal structures and distinctions in the molecular compositions with such structures." @default.
