- W2913066425 abstract "Atrial fibrillation (AF) often occurs in settings of elevated blood pressure, which causes hemodynamic pressure and volume overload of the atria resulting in pathological stretching of myocytes. Distinct atrial regions subjected to highest dilation are proposed to develop the most significant structural and functional remodeling, forming arrhythmogenic ectopic foci that trigger AF. Specialized cardiomyocyte membrane structures, caveolae, are believed to contribute to cardiac mechanosensing via activation of caveolar mechanosensitive chloride channels ClC-2 and ClC-3. We studied 8-weeks post-myocardial infarction heart failure (HF) mice which showed a significantly increased susceptibility to pacing-induced AF (4/4 in HF vs. 1/6 in wild type (WT) mice) and enhanced ectopic activity originated from inter-caval region (ICR) of the right atrium. In WT mice, RT-qPCR analysis of caveolae-localized ClC-2 and ClC-3 revealed significantly higher mRNA expression in ICR and right atrial appendage (RAA) vs. left atrial appendage (LAA). HF resulted in significant downregulation of ClC-2 (33% and 45%, P<0.05 vs. WT) and ClC-3 (38% and 54%, P<0.05 vs. WT) in ICR and RAA, respectively, but not in LAA. Compared with RAA and LAA, ICR experienced the highest (P<0.01) membrane tension calculated as a membrane convolution index from electron microscopy photographs, and showed a more significant downregulation of caveolae structures, while no changes in caveolae scaffolding protein caveolin-3 mRNA level were observed. Pro-fibrotic transforming growth factor beta (TGF-β) mRNA level increase was the highest in ICR and RAA vs. LAA. In HF, ICR showed the highest level of interstitial fibrosis (32±1%) compared with both RAA (18±1%, P<0.001) and LAA (21±2%, P<0.05). Our findings demonstrate thatstretch-induced disruption of caveolae decreases mechanosensitive chloride channel expression and stimulates fibrogenesis which forms arrhythmogenic ectopic foci in vulnerable atrial regions." @default.
