BsAbs can be divided into “IgG-like” or “non-IgG-like” formats based on their resemblance to the native IgG. Several parameters must be considered prior to evaluation of BsAbs in animal models, including antibody cross-reactivity, immunogenicity, pharmacokinetics, and possible interactions with the host immune system. Selection of an appropriate rodent efficacy model is critical to BsAb discovery, development, and translational strategies. For anti-cancer drugs, the transgenic knock-in mouse model is an informative tool to evaluate antibodies that exhibit poor or no cross-reactivity to the murine ortholog of the antigen target. After initial preclinical evaluation, successful lead BsAbs with desirable safety and efficacy properties are further evaluated in clinical studies.
