- W2914312898 abstract "The alkaline transition of monomeric cytochrome c (Cytc) has been studied for decades and has been used as a model system to study protein conformational change. More recently, it has been suggested that monomeric Cytc alkaline conformers may play an important role in regulating initiate the intrinsic apoptotic pathway. Structural and biochemical studies have shown that an available coordination site on the heme iron of dimeric Cytc, leads to the enhancement of the peroxidase activity. We have recently solved a human dimeric Cytc alkaline conformer crystal structure near pH 10. Unlike previously reported structures of either horse or yeast iso-1 Cytc dimers with H2O/OH bound to the heme, the human dimeric Cytc alkaline conformer contains Lys79-heme ligation. It is known that strong Lys-heme ligation blocks peroxidase activity, indicating that the human dimeric Cytc alkaline conformer possesses significantly lower peroxidase activity. We hypothesize that the conformation of dimeric Cytc is pH dependent conferring the ability to tune its peroxidase activity. To test this hypothesis, we are performing a variety of complementary biophysical methods to investigate the alkaline transition of dimeric Cytc. Using UV-Vis spectroscopy, we are investigating the thermodynamics of the alkaline transition. Using stopped-flow methods, we are investigating the kinetics of the alkaline transition and the peroxidase activity. Using EPR spectroscopy, we are probing different populations of high-spin and low-spin heme species across the pH range 6 to 11. We observe that the alkaline transition of dimeric Cytc involves equilibria among three species. As pH increases Met80 coordination is replaced with as with the standard alkaline transition. However, a high-spin species, presumably H2O coordination, is also replaced by Lys coordination. The transition from the high-spin species to Lys coordination appears to be more facile than the transition from Met80 to Lys coordination." @default.
