- W2915003131 abstract "Phosphatidylinositol (4,5)-bisphosphate (PI(4,5)P2), a phospholipid component of cell membranes, plays important roles in many cellular processes. Transitions between non-clustered and clustered states are necessary for the lipid to carry out its multiple roles in a context-dependent manner. Among many factors that influence clustering, specific ion effects have recently begun to be explored. Given that the headgroup is highly negatively charged at physiological conditions, the cation-headgroup interaction is essential for screening the negative charge. The electrostatic interaction is significantly influenced by interaction with other lipids. The present study examines the roles of cholesterol in the PI(4,5)P2 cluster formation via computational and experimental approaches. All-atom molecular dynamics simulations were conducted on pure PI(4,5)P2 and PI(4,5)P2/cholesterol mixed monolayers with different ion compositions and the trajectories were analyzed using graph theory. The trajectories show that cholesterol forms hydrogen bonds with oxygen atoms on the phosphate diester group and the protonated phosphate monoester groups attached to the PI(4,5)P2 headgroup. This leads to the redistribution of ions at the membrane surface and the reorganization of connectivity among PI(4,5)P2 headgroups. Graph-theoretic analysis reveals that cluster size distributions and structural properties (i.e., degree distributions and clustering coefficients) are different between the pure PI(4,5)P2 and PI(4,5)P2/cholesterol mixed monolayers. The clustering trends in the simulations are supported by area/PI(4,5)P2 measurements obtained from a Langmuir trough." @default.
