- W2922140610 abstract "Purpose: N-methyl-D-aspartic acid (NMDA) receptors play an important role in the development of hyperalgesia following inflammation. Recently we found that NR1 receptor subunits were up-regulated following visceral inflammation. However, the role of NR2 receptor subunits is unclear. The NMDA NR2 subunit receptor gene expression may be associated with colonic inflammation. We investigated the regulation of NMDA-NR2 receptor gene expression after TNBS induced colitis in rats. We hypothesized that a transient induced colitis produces modulation of NMDA-NR2 receptors expression in the spinal L5-S1. Methods: Male Sprague-Dawley rats (150 g-250 g) were treated with either 5mg/ml trinitrobenzene sulfonic acid (1.5 ml/kg) (TNBS, Sigma Chemical Co.) in 50% ethanol (n=8) or an equivalent volume of saline (n=3). The agents were delivered with a 24 gauge catheter inserted into the lumen of the colon. The animals were sacrificed at days 2-28 after induction of the colitis and their L5-S1 spinal cord was retrieved for reverse transcription-polymerase chain reaction (RT-PCR) with primers targeted at NR2A, NR2B, NR2C and NR2D. Results: Increased expression of spinal-NR2A and NR2B appeared at 7, 14, and 21 days after TNBS injection compared to saline controls. Conclusion: Selective changes in the expression of the NR2 subunit of the NMDA receptor occur following TNBS induced colitis in the rat. These results suggest a role for spinal NMDA NR2 receptors in the development of neuronal plasticity and resulting visceral hyperalgesia and visceral pain." @default.
