BPA has been detected in human specimens throughout the world and at levels similar to those associated with adverse reproductive outcomes in animal models. Objective: To evaluate associations between urinary BPA concentrations and semen quality parameters.Methods: A prospective, pre-conception cohort of couples without known infertility (women 18–35, men 18–40) was recruited in Utah beginning in early 2012. Recruitment is ongoing. Couples observed cervical mucus to identify the fertile window, during which men and women collected first-morning urine. BPA was measured in each sample using quantitative liquid chromatography-tandem-mass spectrometry. Geometric means (GM) with 95% confidence intervals (CI) for BPA concentrations were calculated. Semen samples were collected through intercourse using Male-FactorPak™ devices and frozen in the participant’s home freezer prior to analysis. Men provided one sample per cycle for up to two menstrual cycles (n=105). Samples were analyzed for volume, total count, concentration, and percent morphologically normal heads and tails. Logistic models were used to estimate the odds of abnormal semen parameters given urinary BPA GM while controlling for age, BMI, income and race.Results: Abnormal concentration (<15 M/mL) was observed in 18 (17%) samples. For every unit increase in BPA geometric mean, the odds of abnormal concentration increased by 1.18 (95% CI 1.01, 1.37). Forty-five (43%) men had morphologically abnormal sperm heads (<30%). Men in the high BPA tertile (=4.67 ng/mL) had a 3.30 (95% CI 1.02, 10.70) increased odds of abnormal sperm heads compared to men in the low BPA tertile (=1.90 ng/mL). No other statistically significant results were observed.Conclusions: These preliminary findings suggest that high BPA exposure is associated with decreased sperm quality." @default.
