- W2940199478 abstract "Insufficient sleep has been linked to an increased risk for cardiovascular disease, but the underlying mechanisms have not been fully elucidated. Endothelial function, an independent predictor of future cardiovascular events and mortality, may be detrimentally affected by sleep deprivation but this has not been tested. Purpose We investigated the influence of total sleep deprivation (TSD) on endothelial function in healthy young men. Methods Twelve men free of sleep disorders, 18 to 27 years of age (22 ± 1 years; mean ± SEM), participated in this study. Endothelial function was assessed using brachial artery flow-mediated dilation (FMD) via ultrasonography in response to reactive hyperemia following 5-min forearm occlusion. FMD was measured at the same time in the morning following a night of: 1) regular sleep (CON) and 2) TSD. Subjects completed TSD under supervision while resting quietly and after abstaining ≥ 20 hrs from caffeine. Results The duration of sleep deprivation ranged from 24.8 to 28.5 hrs (mean: 26.3 ± 1.3 hrs). TSD resulted in a ~42% attenuation in FMD (6.22 ± 1.10 vs. 3.62 ± 0.46 %; P = 0.016; CON vs. TSD). However, brachial artery diameter (4.43 ± 0.11 vs. 4.41 ± 0.11 mm; P = 0.4) and systolic and diastolic blood pressure (117 ± 2 vs. 117 ± 3 mmHg; P = 0.8 and 60 ± 1 vs. 64 ± 3 mmHg; P = 0.1, respectively) remained unchanged. Conclusions Brachial artery endothelial function is attenuated following TSD in healthy young men. Strategies to mitigate the detrimental effects of sleep deprivation on endothelial function may be important for individuals who routinely undergo extended periods of sleep deprivation.
