- W2945919751 abstract "This study was conducted to investigate the role of polyamines (PAs) as priming agents on improving seed vigour and cold resistance with different moisture contents (Mc)(14, 16 and 18%) under storage condition in barley (Hordeum vulgare L.) c.v Usef. Experiments were conducted at the Seed and Plant Certification and Registration Research Institute in 2012 -2014. The seeds were hand harvested at three initial moisture contents as 18, 16 and 14% on wet weight basis. The seed samples then were sealed in polythene bags and stored in conditioned storage and equilibrium for 8 months. Then, seeds were soaked in 20 mg L-1 (w/v) aerated solution of spermine (Spm), Puterscine (Put) and Spermidine (Spd) and distilled water (W) for 16 h at 20±2C°. Seeds dried to initial moisture level in forced air and sealed in polythene bags, stored in a conditioned storage again until one month. Experimental units were arranged in completely randomized design and factorial based on completely randomized design with four replications after harvesting time and one month conditioned storage after priming, respectively. The results indicated that Mc18% improved the germination percentage (GP) and seedling vigour compared to Mc 16 and 14 %. Also after 8 month storage, Mc18% had the highest GP and seedling vigour. The electrical conductivity (EC) decreased in seed by Mc 18% with compared to Mc16 and Mc 14%. PAs treatment improved seed germination, seedling vigor and enhanced anti-oxidative activity. Also, sucrose concentration increased by PAs treatment, whereas fructose and glucose content decreased. Meanwhile, interaction between seed moisture content ×PAs treatment significantly affected on mean of germination time (MGT), sucrose concentration, fructose content, EC, catalase (CAT) and peroxidase (POD) activities in the first year of experiment. Also sucrose concentration, glucose content, EC and POD activities were significantly affected by interaction between seed moisture content ×PAs treatment in the second year of experiment. Optimum moisture content at harvest and the best priming treatment considering to studied traits were 18.0% and priming with spermine. Likewise, in chilling stress condition maximum proline content and seedling vigour were obtained from Put treatment, whereas the lowest electrolyte leakage was recorded for Put treatment. Meanwhile, interaction between seed moisture content ×PAs treatment in cold condition significantly affected on proline content, electrolyte leakage and radicle dry weight in the first year, Also plumule length and radicle dry weight significantly affected by interaction between seed moisture content ×PAs treatment in cold condition in the second year of experiment. Generally, the effectiveness of PAs on improving chilling tolerance and increasing seedling vigor was more pronounced in Put treatment along with sample with 18.0% Mc. Finally there was strong positive relation between antioxidant enzymes (CAT and POD) activity and sucrose concentration in both years of experiment. Likewise, both antioxidant enzymes activity and sucrose content were positivity correlated with final germination percentage and seedling vigor, meanwhile strong positive relation was observed among proline content, seedling growth and PAs treatment in chilling stress condition. It suggests that this technique could apply especially for improving vigor of foundation and registered seeds in barley at the end of storage period or at least one month before of this period because it often shortens the period of storage." @default.
