- W2947521022 abstract "Early development in mammals is the culmination of periods of reading, writing and erasing epigenetic instructions that define and endow fully differentiated gametes with the potential to create new life. The pathways that support these processes are highly dynamic, yet they must also balance the strict enforcement of cell fate decision at critical junctures, to ensure the elaboration of fixed lineages at precise time points. The complex and multilayered information of the epigenome must instruct a single genome in the zygote and orchestrate the differentiation of all subsequent cell types giving rise to the adult organism replete with new germ cells to perpetuate the process in the next generation. The mechanisms and machinery of the epigenetic pathways required in early mammalian development have been studied extensively, especially in the mouse. Here we consider epigenetic regulation required to establish both totipotency and pluripotency, a prerequisite for preimplantation development, leading to embryonic and extraembryonic lineage commitment at the blastocyst stage setting the cellular trajectory for full-term gestation. The processes and mechanistic regulation in these early developmental decisions are critical to ensure the linear direction and high fidelity of cell fate determination. Applying the principles of reprogramming in development to experimental reprogramming in patient-specific regenerative medicine has opened the possibility to form new replacement tissues and organs in the treatment of chronic diseases and to rejuvenate aging pathology extending a longer healthy lifespan." @default.
