The callus induction test was performed using stevia leaf explants on MS medium and hormonal combinations of BAP and NAA (0, 1, 2 and 3 mg/L) with three replications based on completely randomized design and the highest callus fresh weigh (2.69 gr) and callus diameter (1.34 cm) were observed on medium enriched with 2 mg/L BAP + 0 mg/L NAA.Stevia (Stevia rebaudiana Bertoni) is the sweetest plant in the world that is considered as suitable alternative to sugar for treating problems of obesity and diabetes, but its seeds are hard to germinate and most of them are often empty and sterile due to incompatibility phenomenon during their formation. Also, its reproduction by seed prevents homogeneous population to be produced. For this reason, asexual methods and tissue culture are used in order to fast multiplication. In this experiment, Sodium hypochlorite 5% and 2.5% were used for seed sterilization at 5 and 10 minutes. The best disinfection treatment was determined 2.5% sodium hypochlorite at 5 minutes. For seed germination, B5, MS and N6 media with 3 concentrations including full, 1/2 and 1/4 strength were applied and the most germination percentage (66%) was observed in 1/2 concentration of MS medium. Also, micro propagation of single node cuttings was studied in B5, N6 and MS media supplemented with TDZ (0, 0.1, 0.2 and 0.3 mg/L), Kin and BAP (0, 1, 2 and 3 mg/L) based on completely randomized design with 4 replications, and the highest number of shoots (9 shoots) and the longest shoot (13 cm) was obtained from MS medium containing 0.2 and 0.1 mg/L TDZ, respectively. The highest number of roots (53.75 roots) was observed on B5 medium supplemented with 1 mg/L Kin, the longest roots (5.62 and 5.12 cm) on B5 culture medium without plant growth regulators (control) and containing 1 mg/L of Kin, respectively and the highest number of leaves (68 leaves) on B5 medium without plant growth regulators (control). Single node cuttings exhibited pile form in N6 medium and weren’t suitable for Stevia micro propagation. In addition BAP treatment resulted to callus induction. So, N6 medium and BAP were removed from next test. In next experiment, single node explants were cultured on B5 and MS media enriched with Kin (0, 1, 2 and 3 mg/L) and TDZ (0, 0.3, 0.6 and 0.9 mg/L) based on completely randomized design with five replications. In this experiment, maximum number of shoots (12.6 shoots) and leaves (83 leaves) were produced on MS medium supplemented with 0.9 mg/l TDZ and the longest shoots (16.2, 15.8 and 14.8 cm) were related to B5 medium containing 0.9, 0.6 and 0.3 mg/L TDZ, respectively. Maximum number of roots (53 roots) was observed on B5 medium containing 1 mg/L Kin and the longest roots (4.8, 4.7 and 4.7 cm) were related to B5 medium enriched with control, 1 mg Kin and 0.3 mg/L TDZ, respectively. Moreover, the effects of hormonal combinations on single node cuttings growth were assessed on MS and B5.
