- W2947988081 abstract "Colorectal cancer is one of the leading causes of mortality worldwide and there is a need to develop chemotherapeutic agents that induce a specific cell death that is less inflammatory. In this study, the cytotoxic potential of two lactic acid bacteria strains isolated from Philippine traditional fermented foods were investigated against the human colorectal cancer cell line (HCT-116). Initially, cell-free supernatants (CFS) of two LAB ( Lactobacillus plantarum BS25 and Pediococcus acidilactici S3) were prepared with MRS broth, however initial cytotoxicity results via colorimetric assay suggested that MRS can also elicit cytotoxicity effect on the cells. A new protocol for CFS extraction was made using a modified McCoy’s 5A medium containing MRS broth, minimizing the unintended cytotoxic effect of MRS. Gram staining confirmed the homogenous population of each strain grown in the modified medium. It was confirmed via colorimetric assays that the CFS made from the new proposed method of extraction established IC 50 values of 27.819% v/v and 26.174% v/v, from S3 and 4B1, respectively. Moreover, the cytotoxicity that was observed was further validated via morphological analysis. HCT-116 cells characteristically showed membrane blebbing, granulation, and rounding up, which are typical morphological symptoms of cell death, when treated with the CFS from both strains. These results show that the CFS from L. plantarum BS25 and P. acidilactici S3 appear to be potentially promising anti-cancer agents, which can be further purified via bioassay-guided fractionation methods. It was also determined that MRS alone exhibits cytotoxicity against colorectal cancer cells, and a new protocol for collecting CFS for cancer research was successful in excluding the contribution of MRS in causing cytotoxicity on colorectal cancer cells." @default.
