- W2953733144 abstract "In this study we examined the effect of exposing mice to chronic hypoxia and the development of pulmonary hypertension on the responses of isolated pulmonary arteries and aorta to phenylephrine and acute hypoxia. Mice were either exposed to normoxic (21% O2) or hypoxic (10% O2) conditions for 21 days. The contraction to phenylephrine (PE) was markedly inhibited in pulmonary arteries and aorta isolated from hypoxic mice, and this effect of hypoxia was markedly reversed by 1μM catalase. Pulmonary arteries from normoxic mice contracted with 10nM PE show an increase in force and aorta contracted with 100nM PE show a relaxation when exposed to acute hypoxia (PO2 ~10 torr for 20 mins). Catalase markedly reversed the attenuating effects of chronic hypoxia on the relaxation of aorta to acute hypoxia, whereas, catalase did not restore the impairment of contraction to acute hypoxia seen in pulmonary arteries. While catalase did not alter the contraction to 10nM PE in pulmonary arteries from control mice, it enhanced force generation to 10–100nM PE in aorta. Thus, exposure of mice to chronic hypoxia and the development of pulmonary hypertension promote an attenuation of both pulmonary artery contraction and aortic relaxation elicited by acute hypoxia, under conditions where hypoxia attenuates PE-induced force generation through increasing a peroxide mediated relaxation.
