- W2954630763 abstract "Introduction: The role of the c-MET protein is unknown in biliary tract cancer (BTC). ILOGI decided to determine the over-expression of c-MET in patients with BTC and to analyze associations with clinical parameters. Methods: This is a descriptive, retrospective and prospective study. Over-expression was obtained by immunohistochemistry based on intensity and percentage of stained cells, in 58 patients, with assessment: strong positive, weak positive and negative. Overall survival (OS) curves were built using the Kaplan-Meier method in all patients and in subgroups according to stage, gender, tumor origin, and grade of histologic differentiation. The difference in OS between groups was analyzed by the log-rank test. The association between over-expression and grade of differentiation was studied using the chi-square method. Statistical tests were two-tailed with a significant p-value of 0.05. Results: Median age was 58 years; 46/12 samples corresponded to women/men; 52/4/2 to gallbladder cancer/cholangiocarcinoma/undetermined biliary cancer; 4/36/12/6 showed grade of differentiation 1/2/3/not reported. Twenty-nine samples (50%) stained negative for c-MET, 24 (41%) weak positive and 5 (9%) strong positive. Median survival was 18.2, 11.3 and 11.7 months, respectively. The difference in OS between negative and positive (strong and weak) c-MET patients was near statistical significance (p 0.068). In the subgroups of women, grade 1-2, gallbladder cancer and stage III-IV the difference and pattern of survival curves were similar to the overall population. Over-expression correlated with tumor grade (p 0.015), showing an inverse association. Overal survival was similar in patients with grade 1-2 compared to grade 3 tumors, suggesting that the difference in OS between c-MET positive and negative cases is not due to grade of histologic differentiation. Neither response rate nor time-to-progression were associated with c-MET over-expression. Conclusion: Over-expression of c-MET is common in BTC, is associated with the grade of histologic differentiation and could have prognostic value. If the c-MET pathway is important in BTC, inhibitory drugs would have an impact in OS." @default.
