Other tests by using dilutions and employing mechanical mixing with the semen do not indicate whether the spermicide is readily released. Another method called the contact was therefore devised. The apparatus consists of 2 slides each containing a shallow cylindrical well. The well of 1 slide is filled with the test preparation. A drop of semen is placed in the well of the other slide. The slides are then placed together with the semen and test preparation in contact. Air bubbles are avoided. The preparation is then placed in the incubator. Spermatozoa are inspected at regular intervals through the glass slide. No mixing of semen with spermicide occurs. Volumes of semen and test substance remain the same. The preparation is in an enclosed space of known volume. The end point is when no motile spermatozoa remain. Fresh human semen not over 2 hours old produced by masturbation has been used. However it is also important to allow time for liquefaction of the semen to occur before using. Although few tests have been done the method is considered to give consistent and reliable results." @default.
