- W2969868985 abstract "Abstract Parathyroid hormone (PTH) and parathyroid hormone–related protein (PTHrP) act on target organs through one common receptor, PTH/PTHrP receptor type 1 (PTH1R). As an endocrine hormone, the predominant physiologic effect of PTH is to raise blood calcium through actions in the bone and kidney. In contrast, the paracrine-acting factor PTHrP exerts multiple physiologic roles during bone, skin, and breast development. Intracellular signaling downstream of PTH1R primarily occurs via a Gαs/cAMP-linked signaling system. While many membrane-proximal effects of PTH1R signaling have been detailed, it is not yet completely understood how these early signaling events translate into physiologic changes in gene expression in target cells. Here we will review the current understanding of the intracellular signaling systems downstream of PTH1R. With this information, we will review select examples of in vivo PTH1R-dependent actions in bone and kidney. In doing so, we will highlight major knowledge gaps and stress the importance of future studies designed to link detailed knowledge of PTH1R signaling with its actions on endogenous receptors in physiologically important cell types." @default.
