- W2971634218 abstract "2829 Objectives Variability in chromatographic results has been observed in existing miniaturized chromatography systems for Tc-99m disofenin. Our laboratory evaluated various parameters associated with the chromatography procedure in order to identify and correct issues related to the variability observed in chromatography systems. Methods The existing chromatography system utilized to evaluate free Tc-99m pertechnetate in Tc-99m disofenin included ITLC-SA (SA) chromatography strips with 20% NaCl. Using this chromatography system, Tc-99m disofenin remains at the origin (Rf=0) while free Tc-99m pertechnetate migrates with the solvent front (Rf=1.0). Our laboratory evaluated free pertechnetate levels in Tc-99m disofenin for several weeks. For each specific evaluation, Tc-99m disofenin was prepared (50 to 75mCi in a total volume of 4mL) and chromatography performed at 5 and 30 minutes post formulation. For each chromatographic procedure, two specific SA strips were utilized. Following strip development, one strip was cut at the respective cut line and counted, the second strip was scanned using a chromatographic scanner and appropriate analysis performed. Results Chromatographic evaluations of Tc-99m disofenin over a two week interval indicated higher free Tc-99m pertechnetate (>10%) levels on two specific days (Mondays) at 5 minutes post formulation. Strip activitydistributions obtained from scanning demonstrated secondary disofenin radiolabeled components which migrated from the origin past the chromatography cut line, resulting in an apparent higher free Tc-99m pertechnetate level. These secondary disofenin radiolabeled components may be the result of utilizing old generator eluate (48 hours) in the formulation process. Chromatography results at 30 minutes post formulation indicated resolution of these secondary disofenin components, resulting in normal free pertechnetate levels (<5%). In order to minimize the effect of secondary components on the apparent free pertechnetate levels, the strip cut line was changed from an Rf value of 0.6 to 0.7. The chromatography procedure was changed so that the strip was cut immediately following strip development in order to minimize the additional migration of the secondary components. Conclusions Variability in Tc-99m pertechnetate levels in Tc-99m disofenin was investigated. Secondary radiolabeled component were observed insome preparations. Appropriate changes were initiated to reduce these variable effects." @default.
