- W2972804303 abstract "Abstract. Dental caries is a dental disease caused by several factors such as food scraps, bacteria, and tooth. Food scraps that sticks to the surface of tooth will accumulate into plaque and become a good growth medium for bacteria. A large numbers of bacteria that was found is Streptococcus mutans . This study was conducted to determine the active compound group which has a potential as anti-bacterial in red algae-level extract ( Eucheuma spinosum ) using TLC bioautography method. The extraction was carried out by multilevel maceration method with n-hexane, ethyl acetate and methanol. Each extract was tested for anti-bacterial activity by well method on 2%, 4%, and 6% concentration. Results show that inhibitory zone were not formed in n-hexane extract and methanol extract. Meanwhile, inhibitory zone was formed in ethyl acetate extract at each concentration, 7.1 mm; 7.9 mm; and 9.15 mm. The ethyl acetate extract was monitored by TLC using silica gel GF254 stationary phase with ethyl acetate:chloroform (3:2) mobile phase. The testing of anti-bacterial activity was done using contact bioautography method. Results show that there are spots on TLC chromatogram which produce inhibitory zone. Spots characterization was carried out by spotting patches of AlCl3 and it was assumed that spots were flavonoids. Keywords: Red Algae ( Eucheuma spinosum ), Dental Caries, Bioautography TLC, Antibacterial. Abstrak. Karies gigi merupakan penyakit gigi yang disebabkan karena sisa makanan, bakteri, dan gigi. Sisa makanan yang menempel akan bertumpuk menjadi plak dan menjadi media pertumbuhan yang baik bagi bakteri. Bakteri yang ditemukan dalam jumlah besar adalah Streptococcus mutans. Penelitian ini dilakukan untuk mengetahui golongan senyawa aktif yang berpotensi sebagai antibakteri pada ekstrak bertingkat alga merah ( Eucheuma spinosum ) menggunakan metode KLT bioautografi. Ekstraksi dilakukan dengan metode maserasi bertingkat dengan pelarut n-heksana, etil asetat, dan metanol. Setiap ekstrak di uji aktivitas antibakteri menggunakan metode agar sumur pada konsentrasi 2%, 4%, dan 6%. Hasil penelitian menunjukkan bahwa ekstrak n-heksana dan ekstrak metanol tidak terbentuk zona hambat. Sedangkan ekstrak etil asetat terbentuk zona hambat pada setiap konsentrasi sebesar 7,1 mm; 7,9 mm; dan 9,15 mm. Ekstrak terpilih yaitu ekstrak etil asetat dilakukan pemantauan KLT menggunakan fase diam silika gel GF254 dengan fase gerak etil asetat dan kloroform (3:2). Hasilnya menunjukkan terdapat bercak pada kromatogram KLT yang menghasilkan zona hambat. Karakterisasi bercak dilakukan dengan penampak bercak AlCl 3 dan diduga bercak tersebut adalah senyawa flavonoid.
