- W2975842904 abstract "Muscle satellite cell is muscle progenitor cell, which generates myoblasts to regenerate wounded skeletal myofiber; however, the regeneration occurs efficiently with the help of muscle-resident mesenchymal progenitor cells. We reported chemokine CXCL12 and osteopontin from mesenchymal cells beneficial to genetically-modified model mouse carrying dystrophic changes in skeletal muscle. In last WMS congress, we showed the relevance of these factors to Duchenne muscular dystrophy patients. As these results suggest us the factors—muscle relation, in this study, we analyzed the relevance of CXCL12 to skeletal muscle in healthy human. We found CXCL12 plasma concentration was statistically correlated with skeletal muscle mass in healthy persons, which suggests that CXCL12 is an intrinsic modulator of skeletal muscle mass. In determining CXCL12 concentration quantitatively in platelet-removed plasma, we used Quantikine® ELISA kit (R&D Systems, Minneapolis, MN, USA). We measured the concentration according to the manufacturer's instructions. Body composition analysis was carried out by means of InBody ® S10 (InBody Japan, Tokyo, Japan), which measures impedance by applying alternating currents on the human body. Subject lay on spine during measurement. Descriptive statistical analyses were performed with IBM SPSS software ver. 24 (IBM, Chicago, IL, USA). Mean CXCL12 concentrations of healthy males and females were 2.84 ± 0.257 and 2.90 ± 0.257, respectively (mean ± SD ng/mL; healthy male n=26, healthy female n=42). No statistical significance could be detected between male and female. Regression analysis between CXCL12 concentration (x ng/mL) and skeletal muscle mass (y kg) showed y=9.71 + 6.92 × x in male [p=0.02, 95% CI (2.73, 11.11), correlation coefficient r=0.590, p=0.002, n=24]; and y=12.78 + 2.46 × x in female [p=0.04, 95% CI (0.15, 4.77), r=0.576, p=0.02, n=13]. CXCL12 seems a positive regulator with respect to skeletal muscle mass in healthy adults, which suggests that CXCL12 is an intrinsic modulator of skeletal muscle mass. Skeletal muscles' physiological function may depend on CXCL12." @default.
