W2980426110 abstract "Interleukin-33 is a cytokine with pro- and anti-inflammatory properties involved in a number of central nervous system (CNS) diseases including Alzheimer's disease (AD), however its role in these pathologies has not been clearly defined.In order to evaluate the role of the ST2/IL-33 axis in AD we analyzed IL-33 as well as sST2 concentration in the serum and CSF of AD, MCI and HC individuals to explore their possible relation with AD associated inflammation. We analyzed the concentration of IL-33 and its decoy receptor, sST2, in serum and CSF of 30 AD and 30 mild cognitive impairment (MCI) patients, comparing the results to those obtained in 27 healthy controls (HC). Because a dual functionality of IL-33 is known, we also analyzed the concentration of both inflammatory (IL-1β and IL-6) and antiinflammatory (IL-10) cytokines in the same individuals. Results showed that: 1) IL-33 concentration was significantly decreased in serum and CSF of AD and MCI compared to HC; 2) sST2 concentration was increased in serum of AD and MCI but was below detection limit in all CSF; 3) as compared to HC, IL-1β was augmented in sera and CSF of AD and MCI whereas IL-10 was reduced in serum of both AD and MCI; and 4) no differences were observed in IL-16 concentration. In vitro addition of IL-33 resulted in an increased production of IL-10 and in a reduced generation of IL-1β and IL-6 by LPS+Aβ42-stimulated PBMC of all individuals. These data could support the protective role of IL-33 in the pathology and it could suggest IL-33 as a novel therapeutic approach." @default.
