- W2980673669 abstract "African Americans are at higher risk for Alzheimer's disease (AD) than Caucasians. We previously reported that race modifies CSF AD biomarkers, such that African Americans have lower total tau (t-Tau) and phosphorylated tau (p-Tau181), but not Aβ42, in older adults with normal cognition (NC), mild cognitive impairment (MCI), and AD. Importantly, it is unknown if these differences are present in middle-age, a critical period during which common cardiovascular risk factors and asymptomatic AD pathology together emerge. This study aimed to confirm our prior results in a middle age cohort of African Americans and Caucasians with NC, at high risk for AD by virtue of a parental history and overrepresentation of the APOE e4 allele. Participants are from two NIA funded studies: 1) middle-aged African American and Caucasian, cognitively normal subjects with an AD family history (PI: Wharton), and 2) older African American and Caucasian NCs, in a study of NC, MCI, and AD. AD biomarkers (Aβ42, t-Tau and p-Tau181) were measured in the middle-aged cohort using enzyme-linked immunosorbent assays (Fujirebio, Ghent, Belgium), and in the older cohort using Luminex-based multiplex assays (Fujirebio US, Malvern, PA) at Emory. Cross-platform validation studies show strong linear correlations between these two platforms. The older cohort with NC (n=51) was 45% African American, 41% male, 36% APOEe4 positive, and well educated (16.6yrs). The middle aged cohort (n=82; 59.1yrs) included 21 African Americans and was highly educated (85% college graduate). Half of the middle age cohort had at least one APOEe4 allele, consistent with prior studies enrolling participants with an AD parental history. Our main result is that, in both the middle-age and older cohorts, African Americans with NC had lower CSF levels of t-Tau and p-Tau181, but not Aβ, compared to Caucasians. AD brain biomarkers differ by race, in middle-age and older individuals and across the AD process. Middle-aged subjects here represent the third – and youngest – cohort in whom t-Tau and p-Tau181, but not Aβ, differences were found between African Americans and Caucasians. Further research should characterize AD brain biomarkers across race, gender and genetics, and determine when these brain changes begin." @default.
