- W2980820753 abstract "With the aim of increasing the knowledge of the pathophysiology and pathogenic factors emerging at early preclinical Alzheimer's disease (AD) stages, BBRC started the ALFA program for the prospective follow-up of a cohort of cognitively unimpaired subjects. The nested ALFA+ study incorporates deep phenotyping of participants, including blood and CSF sampling, FDG, amyloid and tau PET, MRI, and cognitive testing. It is therefore fundamental to continuously monitor both their adherence and their opinions, needs and satisfaction to ensure maintaining the highest standards and long-term success. We describe here the first 5-year performance, feasibility and participants satisfaction. Baseline visit (2013-14) resulted in the inclusion of 2,743 participants (45-75 years) in the ALFA parent cohort. According to their characteristics, interests and availability for performing study procedures, participants are invited to join observational prevention studies, with different degrees of complexity, and trials. Furthermore, the quality of the procedures and participants’ satisfaction were monitored. Currently, 2,598 subjects are active in the ALFA program (titration rate <6% over 5 years), of which 90.6% rate the ALFA program as excellent. The vast majority reported upfront that they would undergo most of the procedures carried out in ALFA-related projects (MRI [82.27%], LP [78.1%], PET [87.1%]) as well as entering clinical trials (92.31%) (Fig.1). To date, 2,399 MRI scans have been performed, >97% without incidences, being highly valued the attention received through the procedure (Fig.2A). Regarding LP (532 performed) ∼90% of participants reported no adverse events; 5.5/0.2% mild/moderate headache; 3% mild local back pain; 0.5% other mild complications (Fig.2B). Again, the attention received during the LP procedure was excellently valued by ALFA participants, reporting 97% that they would undergo a follow-up LP again." @default.
